Pharmacological Validation

The CA200700 agonist was shown to induce a reporter gene response, in two separate recombinant CHO cell lines expressing either the human β1 or β2 adrenoceptor and a cyclic AMP-responsive secreted placental alkaline phosphatase (SPAP) reporter gene. This response was antagonized by either the b1-selective antagonist CGP 20712A in the human β1 cell line, or the b2-selective antagonist ICI 118551 in the human β2 cell line, respectively. The cyclic AMP-induced expression of SPAP was measured under basal and isoprenaline-stimulated (maximal) conditions.

To determine the KD for CA200700 at human β1, β2 and β3 receptors, radioligand binding was used. Cells were treated with varying concentrations of CA200700 in the presence of a fixed concentration of the radioligand [3H]-CGP 12177. The KD was calculated from the radioligand displacement IC50 value using the Cheng-Prusoff equation.
